Our journey on ALTA3263 clinical trial (tumors with KRAS mutations)

Unlike some older KRAS drugs that target only one mutation (such as G12C), ALTA3263 was designed to inhibit multiple KRAS mutations and may potentially cover more than 90% of KRAS variants.
* It is currently being tested alone and in combination with cetuximab.

Is anyone on this trial? What kind of side effects are you having? How long are they lasting?

At this point, ALTA3263 is still in Phase 1 testing, so there is very limited human side-effect data publicly available. The trial's primary purpose is to determine safety, tolerability, and dosing.

Based on experience with other KRAS inhibitors, oncologists will likely be watching closely for:

Diarrhea
Nausea
Fatigue
Rash or skin changes
Elevated liver enzymes
Decreased appetite
Blood count abnormalities

@mcharlesfrancis I believe it is in late stage 1 testing. They started my husband on 300mg 2x a day. He's only been on it 5 days and has started to get occasional diarrhea, nausea, and fatigue. All seem to be controllable.

Husband is on this trial now. Anyone else? Would like to compare side effects.

What kind of trial is this never heard of it

It's for pancreatic cancer. It is in phase 1. also targets KRAS mutations. Sounds very promising. My husband is tolerating it well. Has his first CT next Tuesday. Fingers crossed!

It is also a dual ON/OFF-state inhibitor, meaning it can bind to KRAS whether it is in its active or inactive form, potentially providing more complete and longer-lasting inhibition of the cancer-driving protein.

Because this is a Phase 1 trial, patients should understand that the primary objective is to establish safety and dosing—not to prove effectiveness. Husband has been on the ALTA3263 trial for 6 weeks now. Had his first CT scan and it was very good! EVERYTHIG is shrinking. woohoo
