Hi @mnmimi, this is an excellent question. I believe from an earlier discussion, that you and your sister will be going to Mayo Clinic in Rochester.
You may wish to read the recommendations provided in this related discussion:

Most of the hotels around Mayo Clinic offer accommodations for a variety of accessibility needs. Wheelchairs are available almost everywhere as are wheelchair entrances, ramps, lifts into pools etc. Many facilities offer kitchens and kitchenettes.

Mimi, I'm so glad your sister got an appointment. When will you be going?
She is there now.
@mnmimi I think a good first step is to check with Concierge Services as @colleenyoung suggested. Every office and person I have worked with at Mayo Clinic has been kind and helpful. Concierge Services is at no cost to you and they will be most helpful.
I can offer my experiences with hotels near Mayo Clinic in Rochester.
All of the hotels I have stayed in are accessible. If a person wishes to have a room that is fully accessible as you've mentioned then that should be specified when making a reservation. The hotels closest to Mayo Clinic tend to be the most expensive. Here are my suggestions based on our experiences:
1) Marriott Residence Inn. There are kitchens in all of the rooms and the Residence Inn at Mayo Clinic connects at the basement level with the extensive subterranean tunnels to and from Mayo Clinic.
2) Marriott TownePlace Suites at Mayo Clinic. Kitchens in all the rooms. They have a very nice buffet breakfast in the morning. This hotel is very close but it is not connected underground with Mayo Clinic.
3) Aspen Suites Hotel. Kitchens in all the rooms. This hotel is across the street from St. Mary's Hospital. It is about a mile from Mayo Clinic however there are accessible shuttles that are very convenient.
There are other hotels that are within a few blocks of Mayo Clinic and within the downtown areas that we have not stayed in. These hotels all offer accessible shuttles.
I do not recommend the Kahler Inn and Suites because of their poor procedures and policies concerning COVID safety.
I hope this is helpful to you and your sister.
If you need a roll-in shower and raised commode with bars on the walls, and a room that easily allows wheelchair access, stay away from the new Hyatt House on Civic Center Dr. Their version of a handicapped-accessible commode is an elevated portable seat extender that sits on top of a regular toilet. There are no bars on the walls. The shower has a small ledge and there is no room to navigate around the furniture in a wheelchair...And did I mention that the only chairs are for the Hi-Top table, meaning that a person has to climb onto the chair and sit at a hi-top table to eat?
I am not in a wheelchair but do need a real elevated commode with stable bars on the walls to help me get up. Horrible time with their set-up.
My husband actually went out a BOUGHT a chair that I could sit on and the manager loaned us a regular-size table to bring into the room so that I could sit at a table.
The staff was very nice but limited in what they could do. I'm not sure who the Hyatt specialist in handicapped accessibility is, but they need re-training! We notified the MAYO concierge service of these problems before we left.
