I tried the Prohealth NMN supplement , and it made me extremely tired . I took the lowest possible dose for 2 weeks , but it felt like I was taking Benadryl . If you look at conversations regarding NMN on Reddit , you will see complaints of fatigue like I experienced , and people that receive increased energy . So I guess it’s a crapshoot .
Lastly , I started questioning if it was a good idea to take it, even if it made me feel great , since I’m on Brinsupri . There’s no way of knowing how the 2 could interact .

@cholash I agree with you. Being careful with supplements is what I'm going to do. Our doctors cannot really guide us here, unless it is a very well-known and understood supplement and there is knowledge of any drug interactions. We're all in new territory here, because Brinsupri is so new. One example of drug interactions: My ID doc said that taking Rifampin or Rifabutin will cause a 35% decrease in the efficacy of Brinsupri. How that was ever determined, who knows? He was reading off his computer program. Well, I decided to take them both, but I have no idea if it's just the Brinsupri being dampened or if it's a two-way street and the Rifabutin efficacy is also impacted while combining these drugs. But at least the doctors know what I'm on.

@ursala7 I'm really curious about the source of that information, since people taking the antibiotics for MAC were excluded from the clinical trials, and post-release information gathering is ongoing. If it is a "test tube" finding, I would likely ignore it until new research is actually published.

@ursala7 rifampin does the same with opiates. Put me into instant withdrawal thanks to my doctor. Rifampin effecting liver enzymes responsible for metabolism of certain drugs (speeding up) the clearance = less effective

@damcipro Wow, not a good scenario. Must have been awful for you.

I read a few clinical human studies on NMN, not sure what's on reddit. NMN is a form of Vitamin B, a derivative of B3. My experience was in line with the results seen in the clinical human studies. If someone has plenty of NAD in their system, additional supplementation does nothing-that was confirmed in human trials but maybe it does the reverse? I don't know about Reddit or even Facebook, but I do try to read clinical human studies. I'm certainly more concerned about Brinsupri which effects neutrophils. Neutrophils are scavengers for abnormal cells and they are immune cells, and on top of that, who did the research? We may not know for at least another 5 to 10 years or more if there are other more serious risks. I asked my NJH ID doctor if taking Brinsupri could affect my body's ability to keep the MABC at bay, which is what I've been doing since 2019, and her response was "we just don't know".

@kathyjjb Your doc is correct, since those with active infection were excluded from the clinical trials. So in effect, those with NTM are effectively a Phase IV trial. I hope someone will be conducting a comprehensive study and reporting on it.

I believe the BEST-BE Study includes those with NTMs which will help. I haven't heard when this will start. I know of 2 large population studies that are in the works (BEST-BE and a European Study), neither are exclusively NTM and Brinsupri but hopefully will include all those taking Brinsupri. The long-term effects that I'm also concerned about are those diseases that neutrophils could help abate. These are the primary reasons why 10 mg is more appealing to me, but I do plan on staying on the 25 mg until my next CT in April 2027.
