Our journey on ALTA3263 clinical trial (tumors with KRAS mutations)

Hello, my mom has been on Alta3263 since January. Tumor has shrunk and been holding stable. Main side effect has been nausea that seems to be increasing over time. She’s also getting more and more lethargic even though CT’s show cancer is stable. Unclear if it is a side effect. She’s repeating CT every 6 weeks. Next one is this week. Thanks for sharing! It is very encouraging that your mom's liver lesion has shrunk and remained controlled on ALTA-3263. That's wonderful to hear.

The increasing nausea and lethargy/fatigue are definitely worth discussing with the trial team. With an investigational drug, I would make sure they know exactly how much these symptoms have changed rather than assuming they are something she simply has to tolerate. The team may be able to adjust supportive medications, look for dehydration or electrolyte problems, or determine whether a dose modification or treatment break is appropriate under the trial protocol.

I would also ask them to check whether something besides the medication could be contributing—such as anemia, dehydration, nutrition, liver function, thyroid problems, infection, or other medications.

Has anyone else currently taking ALTA-3263 experienced increasing nausea or fatigue over time? If so, what has your trial team done that helped?

@mcharlesfrancis thank you for your response! All her symptoms are logged and discussed but generally the response is that the medication is working and everyone is hesitant to modify dosage if it is working, but she can stop it anytime it becomes intolerable. She’s not there yet. She has regular bloodwork to check items you have listed. Results are stable in general, her lethargy is not aligned with test results but she can’t shake it off. CT tomorrow and if all is stable still, will push harder on Dr for suggestions. Thank you, appreciate the support.

@tomrennie Hello, she’s on anti nausea meds, but unsure of the names honestly. Usually she needs to sleep through the morning and then better in the afternoon. But her energy level is way down and nothing is helping with that except the grandkids 🙂

@tomrennie CT is the same, which is good. Overall, tumor shrunk around 50% from start of treatment in January! Dr appointment tomorrow and will discuss symptoms.

After 8 weeks he ended up in the ICU with major lung inflammation. Doc thinks it’s from the trial drug. He’s been here for 19 days. He’s getting better, but it’s a very slow process. Need your prayers……..
