How long before benefits of Camzyos were realized?
I just started the drug 4 days ago.
My symptoms steadily declined over the last three months. Then they really worsened in the last couple of weeks: shortness of breath, light-headedness, tightness or pressure in the chest. These occurred EVERY time I got up from a resting position. I was a little better in the mornings, but got worse as the day progressed.
If my OHCM problem had not been diagnosed, I would have imagined I was about to have a heart attack.
I would like to hear how long others waited before benefits kicked in.
My cardiologist and drug reps say 4 weeks. Anybody realize benefits sooner than that?

Yes, within the first week a very noticeable improvement!! I’m so thankful it was prescribed for me. Have been taking it since May and feel so much better.
My chest pressure stopped in a week. My septum diameter went down in only 8 weeks from 2.2 cm blockage to 1.6 cm. I started to walk 2 miles daily and also ate a lot less food than before adding more vitamins so I am doing great now.

Here's my update: After 10 days on the drug and after a disastrous stress echo, my cardiologist, in consultation with CAMZYOS, stopped the drug. My cardiologist wants to search for and examine other possible problems not fixable with CAMZYOS.
During those 10 days, I experienced NO improvements in labored breathing, pressure/tightness in chest, and increasingly frequent lightheadedness. I'm winded after walking 50 feet on flat terrain.

I just had a catheterization two days ago (11/08/24). The cardiologist surgeon went up the arteries on both sides of the groin to get a complete picture of all arteries in the heart. Good news: all were clear, so no stent was inserted (which would have been done if needed).
The aortic valve also looked good which I had replaced via a TAVR in June 2022. However, the doctor found possible problems with the mitral valve—regurgitation. My next diagnostic step is a TEE—transesophageal echocardiogram. As the name suggests, a thin, flexible tube is put down the throat. A TEE can provide clear images of the heart and all its valves because the transducer is close to the heart and the sound waves don't have to pass through skin, muscle, or bone.
I also wore a heart monitor for two weeks which showed possible electrical problems. So, I may need a pacemaker. But the need for a pacemaker will not be addressed until after the mitral valve problem is addressed.
I would not be surprised if I am put back on CAMZYOS after the dust settles.

Can anyone explain to me how—the mechanism—by which CAMZYOS reduces the diameter of the septum?
Here’s a simplified overview of CAMZYOS's mechanism in reducing septal thickness:
Camzyos belongs to a class of drugs called cardiac myosin inhibitors. Camzyos' mechanism of action (the way it works) is to block the activity of a certain protein that helps your heart muscle contract (squeeze). By blocking this protein, Camzyos relaxes your heart muscle.
Myosin Inhibition: CAMZYOS selectively binds to cardiac myosin, decreasing the interaction between myosin and actin filaments within heart muscle cells. Myosin and actin are crucial for muscle contraction, so by reducing their interaction, CAMZYOS decreases the force of contraction in the heart muscle.
Decreased Hypercontractility: In hypertrophic cardiomyopathy, the heart muscle tends to contract too forcefully, leading to increased workload and progressive thickening of the septum. By reducing the force of contraction, CAMZYOS allows the heart muscle to relax more effectively, which reduces the abnormal pressure and stress on the heart muscle.
Reduction in Septal Thickness Over Time: By decreasing the excessive contraction, CAMZYOS helps to "unload" the heart, leading to a gradual reduction in septal thickness. This happens because the heart no longer has to work as hard to pump blood, allowing the septum to return toward a more normal size.
Improved Blood Flow: With the septum's reduced thickness, CAMZYOS alleviates the obstruction in the left ventricular outflow tract, making it easier for blood to flow out of the heart and reducing symptoms like shortness of breath, chest pain, and fatigue.
In summary, CAMZYOS reduces septal thickness by decreasing the heart's excessive contractility, relieving the stress on heart muscle cells, and allowing the septum to reduce its diameter thickness and resizing to an smaller diameter.
I have HOCM with a heart murmur since birth, I have been told from my echoes that they the gradient is anything from 23 to 121. Has anyone else experienced that echocardiogram including with Stress are really undependable and seeing you seem to get wildly different results? also I don’t have the canning of the septum. I have a genetic small knuckle that sticks out and blocks the outflow valve so I’m not a candidate for surgery. I’ve been debating about Camzyos for several years now the last time I was told my gradient wasn’t bad enough but on December 18, I’m going to have another Stress Echo to determine if that last one was correct or not.
I have a murmur and obstructive HOCM too plus a leaky valve. I am 64 years old, my gradient was between 45 and over 240 mm Hg. So I started Camzyos 7 months ago and my gradient reduced now to less than 45 mm Hg. My septum basal diameter reduced from "2.2 cm" (yes centimeters not millimeters) to 1.6 cm. I am feeling a lot better. I hope you can soon qualify to use Camzyos which is the only medicine that works for me. Good luck and I hope you improve your heart conditions.
