Hello, I’m new to this group. Does anyone know about “conolidine?

Posted by elliptical @elliptical, May 10 12:33pm

I’ve had chronic nerve pain in both legs for 12 years. I don’t have diabetes but doctors ( including Mayo doctors) don’t have an answer.
Gabapentin is the only thing that helps slightly.
I’ve recently heard that products containing conolidine might help but I’m having trouble finding a good source.
Has anyone tried a product? Does it help for nerve pain?

Hello @elliptical, welcome to Connect. There is another older discussion on conolidine that might be helpful to scan through - Conolidine has not helped me at all for peripheral neuropathy: https://connect.mayoclinic.org/discussion/conolidine-has-not-helped-me-at-all-for-peripheral-neuropathy/.
@jetsetter081268, @lorirenee1 and others may have some experience to share with conolidine. While you wait for others to respond, I thought I would share the following references on conolidine.

--- 2011 - Compound offers pain relief without the complications: https://www.nature.com/articles/news.2011.313
--- 2021 - The natural analgesic conolidine targets the newly identified opioid scavenger ACKR3/CXCR7: https://www.nature.com/articles/s41392-021-00548-w

Have you discussed the product with your doctor or pharmacist to see if they have any suggestions?

Sorry, I don’t have any experience with conolidine and am not aware of any information that Mayo recommends it.

does anyone have any knowledge of legitimate research on this medication? I read an article by a Doctor Elizabeth Frost but have not been able to find more

Here is an excerpt from "Anesthesiology and Pain Medicine"...
"Research on conolidine is limited, but the few studies currently available show that the drug holds promise as a possible opiate-like therapeutic for chronic pain. Conolidine was first synthesized in 2011 as part of a study by Tarselli et al." You can find the full test here:
https://www.ncbi.nlm.nih.gov/pmc/articles/PMC8908788/

For a deeper dive, I took a look at this article,
"From Plant to Chemistry: Sources of Active Opioid Antinociceptive Principles for Medicinal Chemistry and Drug Design" (https://www.mdpi.com/1420-3049/28/20/7089) which reviewed over 2 dozen plants with various analgesic and/or opioid effects. Many of these are used in traditional medicine, alone or in combination. Here is part of what they reported about conolidine.
"Conolidine (71, Figure 31) isolated from the bark of the flowering shrub Tabernaemonta divaricata is mainly employed in traditional Chinese and Thai medicine for pain and fever treatment. It contains different alkaloids that possess a carbon-based framework similar to opioids."
The report also goes on to say that the mechanism of action is not yet well understood, and scientists are trying to synthesize forms of conolidine that will be more stable and more effective. When I read those words, I immediately think "not quite ready for prime time" - far too many promising cures and treatment have fallen short of their initial promise.

What does this mean for us?
Someday conolidine, or perhaps another of the compounds being studied, will be another tool in the arsenal of non-opioid pain management.
It may be helpful now, but there is no standardization of these substances, no testing to be sure what you think you are buying is what it says. So...to me that means not to buy from an anonymous promoter on a Website or elsewhere online...but that's just ther skeptic in me.
Conolidine is used in traditional Ayurvedic and Oriental Medicine, so if you have access to a good practitioner, and they have access to a pure supply of bernaemonta divaricata, or crepe jasmine, you might want to try it under their guidance.

I was surprised, with recent ads I have seen, that there wasn't more research around.

Your reaction is the same as mine. I am a retired surgeon and have the same skepticism . I managed to communicate with Elizabeth Frost who published on this medication. I might give it a try and let you know what my experience is

Hi! I tried it for a month, as I’ve tried so many supplements and medications just like others on here.
It did nothing for me. I returned empty bottle and whatever else I bought from GDR Labs with no problem. My credit card was reimbursed quickly.
Good luck.
